چکیده مقاله:

Osteosarcoma is the most frequent malignant bone tumor with a peak incidence in the second and third decades of life. Diagnosis of patients with osteosarcoma is limited to clinico-pathological parameters whereas molecular markers of tumor initiation and promotion have not yet been identified. We aimed to study the expression of the anti-apoptotic survivin gene in osteosarcoma specimens. A total of 35 formalin-fixed, paraffin-embedded (FFPE) specimens including 25 tumoral and 20 non-tumoral paired-margins were collected. The expression of survivin was evaluated by employing real-time PCR and immunohistochemistry. Our data showed that survivin is differently expressed in osteosarcoma samples rather than non-tumoral margins. Real-time PCR showed that survivin is up-regulated in osteosarcoma specimens compared to margins (p<0.01). Furthermore, IHC confirmed that the survivin protein is significantly found in malignant parts of paraffin blocks. Our data show that survivin expression is modulated in osteosarcoma and may be considered as a diagnostic marker for further studies. Detection of Survivin Gene Expression in Formalin-Fixed Paraffin-Embedded Tissues of Human Osteosarcoma: Its Potential Usefulness in Diagnosis and Prognosis of Bone Tumors

Osteosarcoma is a relatively uncommon malignancy however, it is the most common form of primary malignant bone tumors in human. Diagnosis and prognosis of patients with osteosarcoma is limited to clinico-radiopathological parameters, whereas molecular markers of tumor aggression have been poorly identified. Survivin, an inhibitor of apoptosis (IAP), is unique for its expression in human tumors ...

Diagnostic value of direct immunofluorescence of mucosal and skin biopsy specimens on formalin-fixed paraffin-embedded biopsy specimens compared to specimens placed in normal saline as the gold standard method in the diagnosis of a group of autoimmun

Background and Aim: Direct immunofluorescence (DIF) is represented as a gold standard method in diagnosis of autoimmune blistering dermatoses. Normal saline, liquid nitrogen and michel’ solution are a widely accepted media for preserving biopsy samples of skin or mucosa before DIF examination. Occasionally clinicians put the biopsy specimen taken for DIF in formalin 10%, occasionally clinicians...

RNA Sequencing of Formalin-Fixed, Paraffin-Embedded Specimens for Gene Expression Quantification and Data Mining

Background. Proper rRNA depletion is crucial for the successful utilization of FFPE specimens when studying gene expression. We performed a study to evaluate two major rRNA depletion methods: Ribo-Zero and RNase H. RNAs extracted from 4 samples were treated with the two rRNA depletion methods in duplicate and sequenced (N = 16). We evaluated their reducibility, ability to detect RNA, and abilit...

Reliable LC3 and p62 Autophagy Marker Detection in Formalin Fixed Paraffin Embedded Human Tissue by Immunohistochemistry

Autophagy assures cellular homeostasis, and gains increasing importance in cancer, where it impacts on carcinogenesis, propagation of the malignant phenotype and development of resistance. To date, its tissue-based analysis by immunohistochemistry remains poorly standardized. Here we show the feasibility of specifically and reliably assessing the autophagy markers LC3B and p62 (SQSTM1) in forma...

The Utilization of Formalin Fixed-Paraffin-Embedded Specimens in High Throughput Genomic Studies

High throughput genomic assays empower us to study the entire human genome in short time with reasonable cost. Formalin fixed-paraffin-embedded (FFPE) tissue processing remains the most economical approach for longitudinal tissue specimen storage. Therefore, the ability to apply high throughput genomic applications to FFPE specimens can expand clinical assays and discovery. Many studies have me...
